El convento fue construido a principios del siglo XVII sobre la casa natal de Santa Teresa de Jesús e inaugurado en 1636. Fundado por la Orden de los Carmelitas Descalzos, el edificio sufrió graves daños durante las exclaustraciones del siglo XIX, lo que llevó a dos importantes restauraciones, en 1888 y 1982.

La orientación del convento y su disposición particular responden a la intención de situar la cabecera del templo en la habitación de Santa Teresa.
