Il Palazzo del Sedile costituisce l'edificio più importante della piazza: è stato realizzato nel 1540 per adempiere alla funzione di sede delle adunanze municipali dell'università cittadina.
L'attuale struttura si deve ai lavori di restauro effettuati nel 1779. Il palazzo è costituito da un grande arco d'ingresso; in alto, ai lati, spiccano due torrini in stile rococò su cui sono collocati, sul primo, una meridiana e sul secondo, un orologio; al centro, sopra l'arco, vi sono le statue di due dei patroni della città di Matera: Sant'Eustachio a sinistra e la Madonna della Bruna a destra.
Fino al 1944 l'edificio ha ospitato gli uffici dell'amministrazione comunale, attualmente costituisce la sede principale del conservatorio. I sotterranei, anch'essi molo interessanti, sono stati utilizzati per la realizzazione di un auditorium destinato a ospitare concerti e manifestazioni di carattere culturale.
